Applications of BBa_K3942003

We used this part as a piece of design for constructions that were packaged into vesicles by the MCF7 cells after nucleofection.

Used constructions:

5’ - psi(short) - IRES - GFP - lacZ -3’

5’ - psi(short) - GFP -lacZ - 3’

5’ - psi(long) - IRES -GFP - lacZ - 3’

5’ - psi(long) - GFP - lacZ -3’

5’ - IRES - GFP - lacZ - 3’

After collecting supernatant with vesicles and adding it to the targeted HT-29 cells. We discovered that within 24 hours only vesicles with 1st construction(5’ - psi(short) - IRES - GFP - lacZ -3’) caused luminescence In the treated line. Which might be caused by the IRES as it adds additional ribosome landing site.

IRES could be used when you need to increase chances of ribosome landing on your mRNA.
